  • Effects of xylitol on gut microbiota structure and metabolism in a dynamic colonic simulator [Elektronski vir] : master's thesis

    Xylitol is a sugar alcohol, used as a sweetener in many different foods. It has mostly been studied for its effects in the oral cavity, but not a lot of research has been done for its effects on ... human gut microbiota. In this Master's thesis, the dose-dependent effects of xylitol (1, 3 and 5 g/L) on the gut microbiota composition and metabolism were studied, using a dynamic gut simulator of the colon microbiota. The concentration of SCFAs was determined with an HPLC system by comparison the retention time with standards, while ammonia was determined with Nessler’s reagent. Absolute quantification of microbial groups was carried out with qPCR. Statistical analysis of the results showed that addition of xylitol causes significant changes in the number of Faecalibacterium, Roseburia, Bifidobacterium, Lactobacillus, Akkermansia and Blautia coccoides. Moreover, gut metabolism was significantly altered upon the addition of xylitol, since the concentration of butyrate increased, and the concentration of acetate and propionate decreased with an increasing concentration of xylitol, when compared with the control. Furthermore, statistical analysis showed a negative correlation between the concentration of xylitol and concentration of ammonia. The concentration of SCFAs can be explained by cross-feeding mechanisms. Based on the results we can conclude that xylitol shows prebiotic effects on gut microbiota by increasing the yield of beneficial metabolites, especially butyrate. The regulation of the interaction among xylitol, gut microbiota and metabolic activity needs further research
