  • Theoretical description of particle sedimentation in blood considering hematocrit : a 2nd generation mathematical model

    Preparation of therapeutic and diagnostic material from blood requires processing to separate the required materials. As blood is a dynamic fluid, processing may affect the contents of the sample, ... depending on the intrinsic parameters of blood and its constituents, as well as of the extrinsic parameters (temperature and setting of the equipment). The protocol is key in obtaining samples of required quality and quantity and mathematical modeling is a convenient tool in search for the optimal procedure. Here we present the up-graded mathematical model for preparation of platelet and extracellular vesicles rich plasma (PVRP) by centrifugation of blood with maximal recovery of platelets that takes into account the effect of hematocrit. The model indicates that optimal time and optimal volume of plasma decrease with increasing effectivity of plasma counter-current to erythrocyte sedimentation, increasing hematocrit and increasing amount of sample in the tube, and increases with increasing extension of centrifuge rotor. We have compared the measured and the calculated volume of plasma at optimized centrifugation time and estimated the parameter ε describing the counter-current of plasma for 6 individual samples from two blood donors. We assessed ε for each sample individually. Next, the model must be validated on a larger cohort of different blood samples.
